Phil Foden scored twice in the first half, with Erling Haaland adding a third to put City in complete control.
United pulled two goals back through Antony and Anthony Martial, but Haaland restored City's three-goal advantage before the break.
Foden completed his hat-trick early in the second half, with Haaland then converting a penalty to complete the rout.
"It was a big win for us," said Foden. "We knew that United were in good form, but we showed our quality today."
"I think this is a big moment in the title race. We've shown that we're the team to beat, and we're not going to give up our title without a fight."
